چکیده

مقدمه: باکتری‌های مایکوپلاسما ژنیتالیوم و مایکوپلاسما هومینیس از جمله پاتوژن‌های مهم دستگاه تناسلی انسان هستند که نقش مهمی در ایجاد ناباروری ایفا می‌کنند. هدف از انجام این پژوهش، طراحی یک واکسن دوگانه شامل اپی‌توپ‌های هر دو پاتوژن فوق و ارزیابی خواص فیزیکوشیمیایی و پتانسیل واکسن در تهییج سلول‌های سیستم ایمنی میزبان بود.
مواد و روش‌ها: با استفاده از ابزارهای بیوانفورماتیکی متعدد، پس از انتخاب پروتئین‌های سطح سلولی دو باکتری، اپی‌توپ‌های سلول‌های B و T مربوط به هر پروتئین تخمین زده شد. سپس اپی‌توپ‌های اختصاصی، غیر سمی، غیر آلرژن و با خاصیت آنتی‌ژنی بالا شناسایی و جهت ساخت واکسن انتخاب شدند. لینکرهای پپتیدی AAY، GPGPG و KK جهت اتصال اپی‌توپ‌های منتخب و لینکر پپتیدی EAAK جهت اضافه نمودن ادجوانت ایمنی‌زای بتادفنسین به ساختار نهایی واکسن مورد استفاده قرار گرفت، سپس ویژگی‌های ساختاری واکسن طراحی‌شده، از جمله ساختار دوم و سوم، قطبیت، حلالیت و داکینگ مولکولی با گیرنده‌های سیستم ایمنی میزبان ارزیابی شد.
یافته‌ها: بر اساس داده‌های حاصل از تحلیل ساختار دوم واکسن طراحی شده، درصد حضور پیچ، صفحات و مارپیچ آلفا حاکی از پایداری و پتانسیل مناسب پروتئین در تشکیل اپی‌توپ بود و همچنین بررسی ساختار سه‌بعدی آن، قطبیت و حلالیت مطلوب پروتئین واکسن را تأیید نمود، همچنین نتایج حاصل از انجام داکینگ مولکولی، قابلیت بالای این واکسن را در برهم‌کنش با گیرنده‌های سلول‌های سیستم ایمنی میزبان نشان داد.
نتیجه‌گیری: واکسن دوگانه چند اپی‌توپی طراحی شده در این مطالعه، پایداری مطلوب و پتانسیل بالایی در تحریک پاسخ سیستم ایمنی میزبان دارد و می‌تواند جهت توسعه واکسن‌های هدفمند علیه این پاتوژن‌ها مورد استفاده قرار گیرد.
